Brain charts for the human lifespan. –Nature 604(7906): 525-533 (2022) “生长发育图表(growth chart)是公共卫生与健康领域的重要工具。它不仅可以揭示儿童发育(如身高体重)的一般规律,为生长发育(如营养摄入情况)评估提供健康参考,而且也可以为各类发育障碍发出预警信号,用于辅助诊断相关疾病,在家庭养育和临床实践中被广泛应用。” –《科学通报》-悦读科学-“彩巢计划”三部曲—测量(工欲效其事, 必先信其器)、队列(一生最好是少年)、理论(青春期的脑中乾坤) 摘要 在过去的几十年里,神经影像已经成为人类脑科学研究中普遍使用的工具。然而,与身高和体重等人体特征的生长发育图表相比[1],神经影像指标尚缺乏类似参考标准以量化个体差异。因此,我们建立了一个交互式的开放资源,通过已有的或将来采集的磁共振成像(Magnetic Resonance Imaging, MRI)数据样本评估大脑形态。为了将这些参考图表建立在现有的最大最广的数据集上,避免由全球人口多样性造成的MRI研究偏差,我们汇总了来自100多个研究中的101457名志愿者的123984次磁共振扫描影像,他们的年龄范围从孕115天到100岁(图1a)。基于大脑形态结构毕生发展的非线性轨迹[2]和变化率来量化大脑组织的体积和其他全局、局部指标的百分位数。绘制的脑图表发现了从未被揭示过的神经发育里程碑[3],在纵向发展评估中表现出很高的稳定性,并具备应对不同研究之间技术和方法差异的稳健性。同时,与非百分位数的MRI表型相比,脑图表百分位数显示出更高的遗传性,并为揭示神经和精神疾病的神经解剖变异模式提供了非典型大脑结构的标准化测量指标。总之,以多种常用神经影像表型常模轨迹作为基准,脑图表在量化个体变异的道路上迈出了重要一步。
